A microtome is a precision instrument used to cut extremely thin slices of material, typically biological specimens, for microscopic examination. The name is derived from the Greek words mikros (small) and tome (cutting). It is an essential tool in fields such as histology (the study of tissues), pathology (the study of disease), botany, materials science, and forensic science.

Historically, early microtomes were manually operated, requiring skilled technicians to carefully advance a sample across a sharp blade. Modern microtomes are often motorized and computer-controlled, allowing for incredibly precise and consistent sectioning—frequently down to just a few micrometers (millionths of a meter) thick. For context, a standard piece of paper is about 100 micrometers thick, making microtome slices nearly twenty times thinner.

There are several different types of microtomes, each suited for specific applications:

• Rotary Microtome: The most common type in clinical labs; it uses a rotary handwheel mechanism to move a sample vertically against a fixed, extremely sharp blade.
• Cryomicrotome (or Cryostat): Used for freezing samples and cutting sections quickly—essential in surgical pathology for rapid diagnosis while a patient is still on the operating table.
• Vibratome: Uses a vibrating blade to cut through fresh or living tissue with minimal damage, making it ideal for neurobiology research.
• Ultramicrotome: Equipped with diamond or glass knives to cut sections measured in nanometers, specifically for use in electron microscopy.

The process of preparing a sample often involves embedding it in a supporting medium like paraffin wax or resin to provide the necessary stability for slicing. The resulting sections are placed on slides, stained with specific dyes to highlight cellular structures, and finally analyzed. Without the microtome, our understanding of the microscopic world—from the architecture of a cell to the progression of cancer—would remain largely invisible.
