Haematoxylin is not something you'd usually find in an art supply shop, even though it can make a rich blue-purple colour. It comes from the heartwood of the logwood tree, but it is best known for what it does under a microscope. In medicine and biology, haematoxylin is used to stain tissue samples so important parts of cells, especially the nuclei, are easier to see. This helps doctors study biopsies and helps researchers look closely at living tissue. It is often used together with eosin in the well-known H&E stain, one of the most common stains in histology.
Haematoxylin has a long and interesting history. It comes from the heartwood of the Haematoxylum campechianum tree, often called the logwood tree. For centuries, people valued it as a dye for cloth because it could produce dark purple, blue, and black shades. Later, with the growth of microscopy in the 19th century, it became especially important in science and medicine.
Today, haematoxylin is mainly used as a stain in histology, the study of tissues. In practice, it is usually prepared so that it works with a mordant, often an aluminium salt, to bind strongly to parts of cells that contain genetic material. Because of this, cell nuclei usually appear blue to blue-purple under the microscope. Other parts of the tissue can then be stained pink or red with another common dye called eosin.
This combination, called the H&E stain, is the most widely used staining method in pathology. It helps doctors spot changes in tissue, diagnose diseases such as cancer, and understand how cells and organs are affected by illness. Without haematoxylin, many important microscopic details would be much harder to see.
Beyond medicine, haematoxylin is also used in botany, zoology, and other areas of biology whenever scientists need to see cell structure clearly. It is a good example of how a natural substance became an essential laboratory tool. So when a pathologist studies a biopsy slide and sees those clear blue-purple nuclei, haematoxylin is often part of what made that view possible.
