Chloroquine is an organic compound primarily known as a medication used to prevent and treat malaria, a potentially fatal disease transmitted by mosquitoes. For decades, it was a cornerstone of malaria control efforts, working by interfering with the parasite's ability to function within red blood cells. It's part of a class of drugs called 4-aminoquinolines.

Over time, malaria parasites in many regions developed resistance to it, which reduced its effectiveness and led clinicians to rely on other antimalarial medicines in those areas. Where it still works, it can remain an important option.

However, chloroquine's therapeutic potential extends beyond malaria. Doctors discovered that it could also be used to manage symptoms of autoimmune diseases like lupus, rheumatoid arthritis, and Sjögren's syndrome. In these conditions, the immune system mistakenly attacks healthy tissues, and chloroquine helps suppress this overactive response, reducing inflammation and pain.

In early 2020, during the COVID-19 pandemic, chloroquine (and a closely related drug, hydroxychloroquine) gained significant attention as a potential treatment for the virus. Initial reports and anecdotal evidence led to widespread interest, but subsequent large-scale clinical trials demonstrated that it was not effective in preventing or treating COVID-19—and that it could even cause serious heart rhythm problems and other adverse effects.

This resulted in considerable controversy, with debates about its efficacy, safety, and the role of misinformation. Regulatory agencies like the FDA issued warnings against its use for COVID-19 outside of clinical trials.

Like many powerful medicines, chloroquine has meaningful risks. Depending on dose and duration, side effects can include stomach upset, headaches, and itching; with longer-term or high-dose use, it can also damage the retina (the light-sensitive layer at the back of the eye), sometimes affecting vision. It can also interact with other medicines and, in some people, contribute to dangerous heart rhythm disturbances.

Today, chloroquine remains a valuable drug for treating malaria (where local resistance patterns allow) and certain autoimmune conditions—though for autoimmune diseases, doctors often prefer hydroxychloroquine because it tends to be better tolerated. It's crucial to understand its limitations and potential risks, and to rely on evidence-based guidance from healthcare professionals rather than internet hype.
